Overview

This combination therapy regimen consists of the CDK4/6 inhibitor palbociclib (PD-0332991) and the chemotherapy agents 5-fluorouracil (5-FU), oxaliplatin, and leucovorin. The regimen was investigated in a Phase I clinical trial (NCT01522989) led by Georgetown University for the treatment of advanced solid tumor malignancies. Palbociclib is a targeted therapy that inhibits cyclin-dependent kinases 4 and 6, leading to G1 cell cycle arrest and preventing cancer cell proliferation. 5-fluorouracil is an antimetabolite that inhibits thymidylate synthase, while oxaliplatin is a platinum-based agent that causes DNA cross-linking and damage. Leucovorin is included to potentiate the effects of 5-fluorouracil. The combination aims to exploit potential synergies between cell cycle synchronization and cytotoxic DNA damage to overcome treatment resistance in advanced cancers.
