Overview
A combination therapy of **palbociclib**, a cyclin-dependent kinase (CDK) 4/6 inhibitor, and **camizestrant** (AZD9833), an oral selective estrogen receptor degrader (SERD), used for the treatment of estrogen receptor-positive, HER2-negative advanced breast cancer. **Palbociclib** inhibits CDK4/6, causing G1 cell cycle arrest in tumor cells, while **camizestrant** binds, blocks, and degrades the estrogen receptor, inhibiting hormone-driven tumor growth. The combination is in phase 3 trials (SERENA-4, SERENA-6) evaluating efficacy and safety in patients who are heavily pretreated, including prior chemotherapy, endocrine therapies, and previous CDK4/6 inhibitors[1][3][4][5][7]. Early studies show favorable safety and pharmacokinetics profiles and promising anti-tumor activity.
