palbociclib + endocrine therapy

Palbociclib is a selective, reversible small molecule inhibitor of cyclin-dependent kinase 4 (CDK4) and cyclin-dependent kinase 6 (CDK6), developed by Pfizer. It is used in combination with endocrine therapy—such as aromatase inhibitors (e.g., letrozole) or the estrogen receptor antagonist fulvestrant—for the treatment of hormone receptor-positive, HER2-negative advanced or metastatic breast cancer. Palbociclib works by blocking CDK4/6-mediated phosphorylation of the retinoblastoma protein, thereby arresting cell cycle progression from G1 to S phase and inhibiting tumor cell proliferation. The combination with endocrine therapy has been shown to significantly improve progression-free survival compared to endocrine therapy alone in this patient population[1][3][4][5][6][8].
