Overview

Palbociclib + ribociclib is a hypothetical combination of two oral small molecule inhibitors, each targeting cyclin-dependent kinases 4 and 6 (CDK4/6). Both drugs are individually approved for the treatment of hormone receptor-positive, HER2-negative advanced or metastatic breast cancer, typically in combination with endocrine therapy such as aromatase inhibitors or fulvestrant. Palbociclib and ribociclib work by selectively inhibiting CDK4/6, thereby blocking cell cycle progression from G1 to S phase and suppressing proliferation of cancer cells. While both agents have similar mechanisms and efficacy profiles when used separately, there is no clinical evidence supporting their combined use together in a single regimen; they are generally considered alternatives rather than complementary therapies[2][4][5][7]. Each drug has been developed independently—palbociclib by Pfizer and ribociclib by Novartis—and both have demonstrated significant improvements in progression-free survival for patients with HR+/HER2– breast cancer[2][7].
