Palifosfamide tris + doxorubicin is a combination chemotherapy regimen investigated primarily for the treatment of soft tissue sarcoma. Palifosfamide tris is a stabilized salt form of palifosfamide, which itself is the active DNA-alkylating metabolite of ifosfamide. It acts by crosslinking DNA strands, thereby preventing cancer cell replication and inducing cell death[3][6]. Doxorubicin is an anthracycline antibiotic that exerts its antitumor effect mainly through intercalation into DNA and inhibition of topoisomerase II, leading to disruption of DNA repair and generation of cytotoxic free radicals[2]. The combination was developed to improve efficacy over standard regimens; however, clinical trials in metastatic or unresectable soft tissue sarcoma did not show improved progression-free survival compared to doxorubicin alone, leading to early termination of pivotal studies[1][3].
