Overview

Palivizumab and motavizumab are humanized monoclonal antibodies (mAbs) developed by MedImmune (AstraZeneca) for the prevention of respiratory syncytial virus (RSV) infection in high-risk infants. Both antibodies target the highly conserved antigenic site A of the RSV fusion (F) protein, neutralizing the virus by preventing virus-to-cell and cell-to-cell fusion. Palivizumab (Synagis) is the first-generation FDA-approved mAb, while motavizumab (MEDI-524) is a second-generation, affinity-optimized version engineered with 13 amino acid substitutions to enhance binding and potency. Clinical trials, such as NCT00316264, have evaluated the safety and immunogenicity of these antibodies when administered sequentially during the same RSV season.
