Pamrevlumab + FOLFIRINOX is a **combination drug regimen** used in clinical trials for the treatment of **locally advanced, unresectable pancreatic cancer** and metastatic pancreatic ductal adenocarcinoma. The regimen consists of **pamrevlumab**, a fully human monoclonal antibody that targets and inhibits **connective tissue growth factor (CTGF)**, combined with **FOLFIRINOX**, a chemotherapy regimen composed of **folinic acid (leucovorin)**, **fluorouracil (5-FU)**, **irinotecan**, and **oxaliplatin**. Pamrevlumab’s mechanism is to block CTGF, which is implicated in cancer progression and tissue fibrosis. FOLFIRINOX destroys rapidly-dividing cells through cytotoxic effects of its chemotherapeutics. Developed by **FibroGen**, this combination was investigated in Phase 3 trials (e.g., LAPIS, Precision Promise) but failed to significantly improve overall survival compared to placebo plus chemotherapy, leading to discontinuation of pamrevlumab’s development for pancreatic cancer[1][3][5][7].
