Overview

Pamrevlumab + gemcitabine + nab-paclitaxel is an investigational combination therapy for pancreatic cancer. Pamrevlumab is a fully human monoclonal antibody that targets connective tissue growth factor (CTGF), a key mediator of fibrosis and tumor stroma formation. By inhibiting CTGF, pamrevlumab aims to reduce desmoplasia and potentially enhance the effectiveness of chemotherapy. Gemcitabine is a nucleoside analog used as an antimetabolite chemotherapeutic agent that inhibits DNA synthesis. Nab-paclitaxel (nanoparticle albumin-bound paclitaxel) is a microtubule inhibitor formulated to improve delivery and reduce toxicity compared to conventional paclitaxel. The combination has been studied in locally advanced unresectable pancreatic ductal adenocarcinoma (LAPC) as neoadjuvant therapy with the goal of increasing surgical resection rates by reducing tumor fibrosis and improving chemotherapy response[1][5][8]. Clinical trials have also evaluated this regimen in metastatic pancreatic cancer[2][3][9].
