Overview

pamrevlumab + pirfenidone is a combination therapy investigated for idiopathic pulmonary fibrosis (IPF). Pamrevlumab is a fully human monoclonal antibody targeting connective tissue growth factor (CTGF), thereby inhibiting a central mediator of fibrosis and tissue remodeling. Pirfenidone is a small molecule antifibrotic agent with anti-inflammatory properties, primarily used to treat IPF by reducing fibroblast proliferation and collagen synthesis. The rationale for combining these therapies is to potentially enhance anti-fibrotic efficacy, as both target distinct yet complementary pathways involved in IPF progression. Clinical studies, including phase 2 sub-studies, found the combination to be well tolerated, with supportive safety profiles, though efficacy data remain preliminary and further studies are needed to establish clear therapeutic benefit[3][5].
