Drug intelligence / Profile preview
MCLA-129 + Bemena (pamvatamig + ensartinib) is a combination therapy under clinical development for the treatment of advanced solid tumors, particularly non-small cell lung cancer (NSCLC). Pamvatamig (MCLA-129) is a humanized bispecific monoclonal antibody targeting epidermal growth factor receptor (EGFR) and hepatocyte growth factor receptor (c-MET), designed to block oncogenic signaling and overcome resistance mediated by c-MET upregulation. Ensartinib (Bemena) is an orally bioavailable, small-molecule tyrosine kinase inhibitor (TKI) that potently inhibits anaplastic lymphoma kinase (ALK) as well as other kinases such as c-MET, ROS1, and EphA2. The combination is being evaluated in Phase I/II clinical trials to assess its safety, tolerability, pharmacokinetics, and efficacy in patients with advanced solid tumors, leveraging dual inhibition of EGFR and c-MET/ALK pathways to combat resistance mechanisms in lung cancer.
