Overview
panitumumab + 5-fluorouracil + mitomycin C is a combination chemo-immunotherapy regimen investigated for the treatment of localized anal squamous cell carcinoma. The regimen combines panitumumab, a recombinant human IgG2 monoclonal antibody that binds specifically to the epidermal growth factor receptor (EGFR), with the antimetabolite 5-fluorouracil (5-FU) and the antitumor antibiotic mitomycin C. Developed by the Grupo Español Multidisciplinario del Cancer Digestivo (GEMCAD) and evaluated in the Phase 2 VITAL trial (GEMCAD-09-02), this combination is administered concurrently with radiotherapy. Panitumumab works by competitive inhibition of ligand binding to EGFR, while 5-FU and mitomycin C act as cytotoxic agents and radiosensitizers, collectively aiming to improve locoregional control in patients with T2-4 anal tumors.
