Overview
This regimen is a combination of four drugs—panitumumab (a monoclonal antibody), oxaliplatin (a platinum-based chemotherapy), folinic acid (leucovorin, a vitamin B derivative used to enhance the effects of fluorouracil), and 5-fluorouracil (a pyrimidine analog antimetabolite chemotherapy). It is primarily used as first-line treatment for patients with RAS wild-type metastatic colorectal cancer. Panitumumab targets the epidermal growth factor receptor (EGFR) on cancer cells, inhibiting cell proliferation. Oxaliplatin forms DNA crosslinks that inhibit DNA replication and transcription. Folinic acid enhances the binding of 5-fluorouracil to thymidylate synthase, increasing its cytotoxicity. 5-Fluorouracil inhibits thymidylate synthase, disrupting DNA synthesis and leading to cell death. This combination has demonstrated improved progression-free survival compared to regimens without panitumumab in clinical trials[1][2][3][6].
