Panitumumab + sotorasib is a combination therapy that targets both EGFR and KRAS G12C pathways. Sotorasib is a selective and irreversible KRAS G12C inhibitor that locks the mutant protein in an inactive state, while panitumumab is a monoclonal antibody that binds to EGFR, blocking its activation and downstream signaling. This dual inhibition approach was developed to overcome resistance mechanisms observed with KRAS G12C inhibitor monotherapy in colorectal cancer.
