Panitumumab + trifluridine-tipiracil is a combination therapy used primarily in the treatment of refractory RAS wild-type metastatic colorectal cancer (mCRC). Panitumumab is a fully human monoclonal antibody that targets and antagonizes the epidermal growth factor receptor (EGFR), thereby inhibiting downstream signaling pathways involved in tumor cell proliferation and survival. Trifluridine-tipiracil (also known as FTD/TPI) is an oral antineoplastic agent composed of two components: trifluridine, a nucleoside analog that incorporates into DNA and disrupts its function, and tipiracil, which inhibits thymidine phosphorylase to increase the bioavailability of trifluridine. The combination has demonstrated improved progression-free survival compared to trifluridine-tipiracil alone in patients with mCRC who are refractory or intolerant to standard therapies other than anti-EGFR therapy[1][2][3][4][5][7].
