panobinostat + bortezomib

Panobinostat + bortezomib is a combination therapy used primarily with dexamethasone (forming the PanBorDex regimen) for treating relapsed refractory multiple myeloma (RRMM). This combination demonstrates synergistic activity against multiple myeloma cells through complementary mechanisms of action. Panobinostat is a non-selective histone deacetylase (HDAC) inhibitor that increases acetylation of histone proteins, inducing cell cycle arrest and/or apoptosis of tumor cells. It also inhibits the aggresome pathway, which may contribute to overcoming resistance to proteasome inhibitors. Bortezomib is a proteasome inhibitor that disrupts protein degradation in cancer cells, leading to cell death. When combined with panobinostat, there is a synergistic cytotoxic effect that enhances anti-cancer activity.
