Overview

This is a **three-drug combination therapy** consisting of **panobinostat** (a pan-histone deacetylase inhibitor), **bortezomib** (a proteasome inhibitor), and **dexamethasone** (a synthetic glucocorticoid corticosteroid). Panobinostat inhibits histone deacetylase enzymes, resulting in increased acetylation of histones and non-histone proteins, altered gene transcription, cell cycle arrest, and apoptosis. Bortezomib blocks the proteasome pathway of protein degradation, leading to accumulation of misfolded proteins and apoptotic cell death in malignant plasma cells. Dexamethasone enhances antimyeloma responses and reduces inflammation. The combination is primarily used for the **treatment of relapsed or relapsed and refractory multiple myeloma** in adults who have received at least 2 prior regimens including bortezomib and an immunomodulatory agent. The combination has demonstrated a **progression-free survival benefit** in clinical trials, leading to regulatory approval in the US and EU for this indication[1][2][3][4][5][8][9].
