Panobinostat + carboplatin + paclitaxel is a combination regimen under investigation for the treatment of advanced solid tumors. Panobinostat is a non-selective histone deacetylase (HDAC) inhibitor that modulates gene expression and protein metabolism by inhibiting class I, II, and IV HDACs. This leads to cell cycle arrest and apoptosis in cancer cells. Carboplatin is a platinum-based chemotherapy agent that forms DNA adducts resulting in DNA damage and apoptosis. Paclitaxel stabilizes microtubules and prevents their depolymerization during cell division, causing mitotic arrest and subsequent cell death. The combination has shown synergistic antitumor effects in preclinical models as well as early-phase clinical trials for advanced solid tumors such as ovarian cancer and non-small-cell lung cancer[1][4][5][6]. The main dose-limiting toxicities are myelosuppression (neutropenia and thrombocytopenia), with other common side effects including diarrhea, fatigue, vomiting[1].
