Overview

PEI 15-216 is a combination therapy consisting of panobinostat and everolimus, developed by the Fundació Sant Joan de Déu for the treatment of diffuse intrinsic pontine glioma (DIPG). Panobinostat is a potent, non-selective histone deacetylase (HDAC) inhibitor that induces cell cycle arrest and apoptosis in tumor cells by increasing histone acetylation and modulating gene expression. Everolimus is an orally active inhibitor of the mechanistic target of rapamycin (mTOR), specifically targeting the mTORC1 complex to inhibit protein synthesis and cell proliferation. The combination aims to synergistically target the epigenetic dysregulation and signaling pathways characteristic of DIPG, a highly aggressive pediatric brainstem tumor. The regimen was evaluated in a Phase I/II clinical trial (NCT02717455) to determine safety and efficacy in pediatric patients.
