Panobinostat + trastuzumab is a combination therapy consisting of panobinostat, a histone deacetylase (HDAC) inhibitor, and trastuzumab, a HER2-targeted monoclonal antibody. This combination has been investigated primarily for HER2-positive metastatic breast cancer patients who have progressed on or after trastuzumab-based therapy. Preclinical studies demonstrated synergistic effects when these agents are combined in HER2-positive breast cancer models. Panobinostat inhibits class I, II, and IV HDAC proteins, affecting the acetylation of proteins involved in various biological processes including DNA replication, chromatin remodeling, and cell cycle progression. Trastuzumab targets the HER2 receptor that is overexpressed in approximately 20-25% of breast cancers.
