Overview
Dolonol is a fixed-dose combination analgesic and antipyretic medication manufactured by Eskay Therapeutics in Ghana. It contains paracetamol (acetaminophen) and caffeine, typically in a 500 mg/30 mg ratio. Paracetamol provides pain relief and fever reduction by inhibiting cyclooxygenase (COX) enzymes, primarily within the central nervous system, thereby reducing prostaglandin synthesis. Caffeine is included as an analgesic adjuvant; it acts as an antagonist at adenosine receptors, which can lead to vasoconstriction of cerebral blood vessels and potentially increase the absorption and effectiveness of paracetamol. Dolonol is used for the symptomatic relief of mild to moderate pain conditions such as headache, migraine, toothache, and musculoskeletal pain, as well as for managing febrile states.
