Overview

Paracetamol + caffeine + chlorpheniramine is an approved, fixed-dose combination small molecule drug used for the symptomatic relief of cold and flu symptoms, such as fever, headache, rhinorrhea, sneezing, and watery eyes. The combination consists of three active ingredients: paracetamol (acetaminophen), an analgesic and antipyretic that acts by inhibiting cyclooxygenase (COX) enzymes in the central nervous system; caffeine, a central nervous system stimulant that blocks adenosine receptors and enhances the analgesic efficacy of paracetamol; and chlorpheniramine (chlorphenamine), a first-generation H1 antihistamine that competitively inhibits histamine H1 receptors and possesses anticholinergic activity to reduce nasal secretions. It is marketed under brand names such as Calmagrip Forte by Pharmex.
