Panadeine Extra is a fixed-dose combination analgesic containing 500 mg paracetamol (acetaminophen) and 15 mg codeine phosphate. It is used for the relief of moderate to severe pain, including postoperative dental pain, where non-opioid analgesics alone are insufficient. Paracetamol provides analgesic and antipyretic effects primarily through the central nervous system, likely by inhibiting cyclooxygenase enzymes and modulating central pain pathways. Codeine is an opioid prodrug that is metabolized by the liver enzyme CYP2D6 into morphine, which then acts as an agonist at mu-opioid receptors to inhibit pain signaling. The product is historically associated with GlaxoSmithKline Consumer Healthcare (now Haleon) and has served as a reference comparator in clinical trials evaluating new analgesic combinations, such as those developed by Reckitt Benckiser.
