Overview

Paracetamol + diclofenac + tramadol is a fixed-dose combination of three analgesic drugs used for pain relief. Paracetamol (acetaminophen) acts as an analgesic and antipyretic by inhibiting prostaglandin synthesis in the central nervous system, thereby reducing pain and fever. Diclofenac is a nonsteroidal anti-inflammatory drug (NSAID) that works by inhibiting cyclooxygenase (COX-1 and COX-2), leading to decreased formation of prostaglandins involved in inflammation, pain, and fever. Tramadol is a centrally acting synthetic opioid analgesic that functions as a μ-opioid receptor agonist and also inhibits the reuptake of serotonin and norepinephrine, modulating descending inhibitory pain pathways. The combination provides multimodal analgesia through complementary mechanisms—peripheral inhibition of prostaglandin synthesis (paracetamol, diclofenac) and central modulation of pain perception (tramadol)—and is indicated for moderate to severe acute or chronic pain when monotherapy is insufficient[1][5].
