Overview

Paracetamol + ketorolac is a fixed-dose combination of two analgesic agents used for the management of moderate to severe pain, particularly in acute settings such as post-operative pain. Paracetamol (also known as acetaminophen) is an analgesic and antipyretic that works primarily by inhibiting prostaglandin synthesis in the central nervous system, leading to reduced perception of pain and fever. Ketorolac is a nonsteroidal anti-inflammatory drug (NSAID) that exerts its effects by inhibiting cyclooxygenase (COX-1 and COX-2), thereby reducing the synthesis of prostaglandins involved in inflammation, pain, and fever. The combination has been shown to provide superior or supra-additive analgesia compared with either agent alone in some clinical studies[1][3]. This effect appears especially relevant for inflammatory or acute nociceptive pain models. No significant pharmacokinetic interactions have been observed between these two drugs when administered together[3]. The combination is generally used short-term due to the risk profile associated with NSAIDs like ketorolac.
