Paradote is a fixed-dose combination pharmaceutical product containing the analgesic and antipyretic **paracetamol** (acetaminophen) and the amino acid **methionine**. It is primarily indicated for the relief of mild to moderate pain and fever. The inclusion of methionine serves as a hepatoprotective strategy; methionine is a metabolic precursor to glutathione, which is the primary endogenous antioxidant required to detoxify N-acetyl-p-benzoquinone imine (NAPQI), the toxic metabolite of paracetamol. In the event of an overdose, NAPQI depletes hepatic glutathione stores, leading to liver necrosis. By providing supplemental methionine, Paradote helps maintain or replenish glutathione levels, thereby reducing the risk of severe liver damage. The drug is distributed by Equity Pharmaceuticals, particularly within their hospital division in South Africa.
