Drug intelligence / Profile preview
paracetamol + morphine + ketoprofen + ketamine is a multimodal analgesic combination regimen being evaluated for the management of acute pain during vaso-occlusive crises (VOC) in adult patients with sickle cell disease. This combination is studied as Group 08 in the PAMAVOC adaptive platform trial (NCT07826988), sponsored by Assistance Publique - Hôpitaux de Paris. The regimen combines paracetamol (acetaminophen), patient-controlled analgesia (PCA) morphine, and ketoprofen, with the potential addition of low-dose ketamine as an off-label co-analgesic for hyperalgic crises. By targeting multiple pain pathways—including cyclooxygenase inhibition, mu-opioid receptor activation, and NMDA receptor antagonism—the multimodal approach aims to optimize pain control while reducing overall opioid consumption and its associated adverse effects.
