Overview

Parecoxib + loxoprofen is a sequential analgesic regimen consisting of an initial intravenous dose of parecoxib followed by a multi-day course of oral loxoprofen. This specific combination was evaluated by Peking Union Medical College Hospital in a randomized, non-inferiority clinical trial for the management of postoperative pain in patients undergoing transnasal transsphenoidal surgery for pituitary adenomas. Parecoxib is a sulfonamide-based prodrug of valdecoxib that acts as a selective cyclooxygenase-2 (COX-2) inhibitor, providing rapid pain relief via intravenous administration. Loxoprofen is a non-selective propionic acid derivative NSAID that inhibits both COX-1 and COX-2 enzymes, maintaining analgesia through oral administration. The regimen is designed to provide effective multimodal analgesia while minimizing the need for opioid medications like tramadol.
