This drug is a fixed-dose combination of four direct acting antivirals—paritaprevir, ritonavir, ombitasvir, and dasabuvir—used with or without ribavirin for the treatment of chronic hepatitis C virus (HCV) infection, particularly genotype 1. Paritaprevir is an HCV NS3/4A protease inhibitor that blocks viral replication by inhibiting polyprotein cleavage. Ritonavir acts as a pharmacokinetic booster by inhibiting CYP3A-mediated metabolism to increase paritaprevir levels but has no direct anti-HCV activity. Ombitasvir is an HCV NS5A inhibitor that interferes with viral RNA replication and virion assembly. Dasabuvir is a non-nucleoside NS5B polymerase inhibitor that impedes HCV RNA-dependent RNA polymerase activity required for viral replication. Ribavirin is a nucleoside analog antiviral used in combination to enhance efficacy in certain patient populations[1][2][6][8]. The regimen was developed by AbbVie and marketed as Viekira Pak or Viekira Pak-RBV.
