Paroxetine is a selective serotonin reuptake inhibitor (SSRI) that works by inhibiting the serotonin transporter (SERT), thereby increasing serotonin levels in the synaptic cleft. Duloxetine is a serotonin-norepinephrine reuptake inhibitor (SNRI) that inhibits both the serotonin transporter (SERT) and the norepinephrine transporter (NET). The trial aimed to compare the safety profiles of these two classes of antidepressants regarding their impact on the autonomic nervous system, which is often dysregulated in depressed patients and can contribute to cardiovascular risk.
