CRx-139 is an oral synergistic combination drug candidate composed of low-dose prednisolone (a glucocorticoid steroid) and paroxetine (a selective serotonin reuptake inhibitor antidepressant). It was developed to enhance the immuno-modulatory activity of a low-dose glucocorticoid while minimizing steroid-related side effects. The combination targets multiple biological pathways simultaneously and is designed for the treatment of immuno-inflammatory diseases, particularly rheumatoid arthritis. Mechanistically, CRx-139 inhibits pro-inflammatory biomarkers such as TNF-alpha, IL-6, and C-reactive protein while increasing anti-inflammatory cytokine IL-10. The drug was developed by CombinatoRx as part of their approach to create novel synergistic pharmaceuticals that act on complex disease biology[1][2][3][4][5].
