Overview

Combination of parsaclisib, a highly selective phosphatidylinositol 3-kinase delta (PI3Kδ) inhibitor, and pembrolizumab, a programmed death-1 (PD-1) inhibitor monoclonal antibody. This combination was investigated in patients with advanced or metastatic solid tumors who had disease progression following all available therapies. The rationale for combining these agents was to enhance antitumor immune responses by targeting both the PI3Kδ pathway in immune cells and the PD-1/PD-L1 checkpoint axis. The combination showed modest clinical activity but did not demonstrate sufficient efficacy to support continued development for solid tumors[2][1].
