Drug intelligence / Profile preview
Pasritamig + docetaxel is an investigational combination therapy being developed for the treatment of metastatic castration-resistant prostate cancer (mCRPC). Pasritamig (JNJ-78278343) is a first-in-class, bispecific T-cell-engaging antibody that targets human kallikrein 2 (KLK2), a prostate-specific antigen, and CD3 on T cells, thereby redirecting T cells to selectively destroy KLK2-expressing prostate cancer cells. Docetaxel is an established taxane chemotherapy that inhibits microtubule depolymerization, leading to cell cycle arrest and apoptosis. The combination is designed to leverage the cytotoxic effects of chemotherapy alongside targeted immunotherapy to improve outcomes in patients whose disease has progressed after treatment with an androgen receptor pathway inhibitor (ARPI). In clinical trials, the combination has demonstrated a manageable safety profile consistent with docetaxel monotherapy and encouraging clinical activity, including high rates of prostate-specific antigen (PSA) response.
