Overview

Patritumab + trastuzumab is an investigational combination therapy consisting of two monoclonal antibodies targeting different members of the human epidermal growth factor receptor (HER) family. Patritumab is a fully human monoclonal antibody that targets HER3 (ERBB3), inhibiting its signaling and potentially reducing tumor cell proliferation and survival. Trastuzumab is a well-established monoclonal antibody that targets HER2 (ERBB2), blocking downstream signaling pathways involved in cell growth and survival. The combination has been studied primarily in patients with HER2-overexpressing metastatic breast cancer, aiming to enhance antitumor efficacy by dual inhibition of the HER family receptors. Clinical studies have shown this combination to be tolerable, with promising efficacy signals such as improved response rates and progression-free survival[1][6][4][2]. The regimen has been evaluated in phase I/Ib trials, often alongside paclitaxel chemotherapy.
