Pazopanib is an oral, multi-targeted receptor tyrosine kinase inhibitor (TKI) that inhibits vascular endothelial growth factor receptors (VEGFR-1, -2, and -3), platelet-derived growth factor receptors (PDGFR-α and -β), and the stem cell factor receptor (c-Kit). By blocking these pathways, pazopanib inhibits tumor angiogenesis and cell proliferation. It is FDA-approved for advanced renal cell carcinoma and soft tissue sarcoma. In the specific context of the New Mexico Cancer Research Alliance, pazopanib is being investigated in a Phase I combination study with carboplatin, a platinum-based chemotherapeutic agent that functions as a DNA alkylator to induce apoptosis in rapidly dividing cancer cells. This combination therapy is being evaluated for its safety and efficacy in patients with advanced solid malignancies.
