Overview

Pazopanib + vinflunine is an investigational combination therapy consisting of two small molecule antineoplastic agents. Pazopanib is a second-generation multitargeted tyrosine kinase inhibitor that targets vascular endothelial growth factor receptors (VEGFR-1, -2, -3), platelet-derived growth factor receptors (PDGFR-α and PDGFR-β), and c-KIT, thereby inhibiting angiogenesis and tumor growth. Vinflunine is a microtubule inhibitor belonging to the vinca alkaloid class, disrupting mitotic spindle formation and cell division. This combination was studied in patients with metastatic transitional cell carcinoma of the urothelium who had failed first-line platinum-containing therapy. In phase I trials, the regimen was poorly tolerated due to significant toxicities such as febrile neutropenia and hepatobiliary disorders; as a result, further development was halted at an early stage[1][2][5].
