PBSC-M87o gene-modified CD34+ hematopoietic stem cells is an investigational autologous or allogeneic gene-modified cell therapy developed by the University Medical Center Hamburg-Eppendorf. The therapy involves the ex vivo transduction of peripheral blood-derived CD34+ hematopoietic stem cells (HSCs) with the M87o retroviral vector. This vector expresses a membrane-anchored C-peptide derived from the HIV-1 gp41 region, which acts as a fusion inhibitor to prevent HIV-1 entry into the cell. The primary objective is to reconstitute an HIV-resistant immune system in HIV-positive patients who are undergoing high-dose chemotherapy and hematopoietic stem cell transplantation for hematologic malignancies, such as lymphoma. By providing a population of cells resistant to productive HIV-1 infection, the therapy aims to control viral replication while simultaneously treating the underlying cancer.
