PCT1:CO-STIM is an autologous T cell therapy in which a patient’s own T cells are genetically engineered to express a specific T cell receptor (TCR) targeting the retinoblastoma tumor-suppressor gene (RB1) protein, which is found in certain cancers such as triple-negative breast cancer and melanoma. The "CO-STIM" component refers to the integration of a proprietary costimulatory signaling domain into the intracellular part of the TCR, designed to enhance and prolong T cell activity within the immunosuppressive tumor microenvironment. This next-generation approach aims to increase both specificity for cancer cells and durability of response by overcoming common mechanisms of immune evasion in solid tumors[6][7][1]. The therapy is being developed by Pan Cancer T BV, based in Rotterdam, Netherlands[6][4].
