**PCUR-101 + abiraterone + prednisone** is a combination regimen investigated for the treatment of metastatic castration-resistant prostate cancer (mCRPC). PCUR-101 is an orally bioavailable synthetic small molecule derived from plumbagin and developed by Pellficure Pharmaceuticals. It acts as an antineoplastic agent and primarily inhibits STAT3, protein kinase C epsilon, and proto-oncogene protein c-akt (AKT) signaling. In clinical studies, this combination aims to enhance androgen deprivation therapy (ADT) effectiveness. Abiraterone is a CYP17 inhibitor suppressing androgen biosynthesis, while prednisone is a corticosteroid used to manage abiraterone-related side effects and may have additional antitumor activity. This regimen was evaluated in early-phase clinical trials but development was terminated after phase I due to trial discontinuation and possibly insufficient enrollment.
