Overview

PD-0360324 + cyclophosphamide is an investigational combination therapy being evaluated for the treatment of recurrent platinum-resistant epithelial ovarian, fallopian tube, and primary peritoneal cancers. PD-0360324 is a humanized monoclonal antibody that targets colony-stimulating factor 1 (CSF1), a cytokine involved in the recruitment and polarization of tumor-associated macrophages (TAMs) that contribute to an immunosuppressive tumor microenvironment. By inhibiting CSF1, PD-0360324 aims to reduce the presence of these suppressive macrophages. Cyclophosphamide is a nitrogen mustard alkylating agent that cross-links DNA, interfering with cell replication. When used at low metronomic doses, cyclophosphamide can also exert immunomodulatory effects, such as the depletion of regulatory T cells (Tregs), potentially enhancing the anti-tumor immune response initiated by anti-macrophage therapy.
