A combination therapy consisting of a programmed cell death protein 1 (PD-1) monoclonal antibody and a histone deacetylase (HDAC) inhibitor. This regimen is under investigation for its synergistic anti-tumor effects in various cancers, including melanoma, urothelial carcinoma, sarcoma, B-cell lymphoma, hepatocellular carcinoma (HCC), and colorectal cancer. PD-1 antibodies function as immune checkpoint inhibitors that block the PD-1 pathway to reinvigorate exhausted T cells and enhance anti-tumor immunity. HDAC inhibitors modulate gene expression by altering chromatin structure through inhibition of histone deacetylases, leading to increased tumor antigen presentation and immunogenicity. The combination aims to overcome resistance to single-agent immunotherapy by enhancing T cell infiltration and promoting apoptosis in tumor cells[2][3][4][5][6][8].
