Overview

PD-1 inhibitors + chemotherapy refers to the combination of programmed cell death protein 1 (PD-1) immune checkpoint inhibitors with standard chemotherapeutic agents for cancer treatment. PD-1 inhibitors are monoclonal antibodies that block the interaction between PD-1 on T cells and its ligand PD-L1 on tumor or other cells, thereby releasing the "brakes" on T-cell activity and enhancing anti-tumor immunity[2][4]. Chemotherapy works by directly killing rapidly dividing cancer cells through cytotoxic mechanisms. The rationale for combining these modalities is to leverage both direct tumor cell killing (chemotherapy) and immune-mediated tumor destruction (immunotherapy), potentially improving response rates and overcoming resistance seen with either therapy alone[3]. This approach is used in multiple cancers including non-small cell lung cancer, melanoma, kidney cancer, head and neck cancers, among others[4]. Commonly used PD-1 inhibitors in such combinations include pembrolizumab (Keytruda), nivolumab (Opdivo), and cemiplimab (Libtayo)[2][3][4].
