PDC*lung01 + pembrolizumab is a combination therapy consisting of PDC*lung01, an off-the-shelf therapeutic cancer vaccine developed by PDC*line Pharma, and pembrolizumab, an anti-PD-1 immune checkpoint inhibitor. PDC*lung01 is composed of irradiated human plasmacytoid dendritic cells (PDC*line) loaded with HLA-A*02:01-restricted peptides derived from multiple tumor antigens (NY-ESO-1, MAGE-A3, MAGE-A4, Multi-MAGE-A, MUC1, Survivin and Melan-A). The vaccine acts as a potent professional antigen-presenting cell to prime and boost cytotoxic CD8+ T-cell responses against tumor cells. Pembrolizumab blocks the PD-1 receptor on T-cells to prevent immune evasion by tumors. The combination has shown promising clinical activity in phase I/II trials for stage IV non-small cell lung cancer (NSCLC) patients with high PD-L1 expression (≥50%), demonstrating improved objective response rates and progression-free survival compared to pembrolizumab alone. The safety profile is mild and consistent with other vaccines[1][2][3].
