PDPHV-201401 is a preventive HIV-1 vaccine candidate developed by Worcester HIV Vaccine (WHV) using a polyvalent DNA-prime/protein-boost strategy. The vaccine is designed to elicit broad immune responses against multiple HIV-1 subtypes, specifically clades A, B, C, and A/E. The DNA component consists of five plasmids encoding envelope (Env) gp120 proteins from clades A, B, C, and A/E, along with a Gag protein from clade C. The protein component comprises four recombinant gp120 proteins matched to the DNA component, formulated with the GLA-SE (glucopyranosyl lipid adjuvant in stable emulsion) adjuvant. Originally developed at the University of Massachusetts Medical School by Dr. Shan Lu, the vaccine has undergone Phase 1 clinical testing (HVTN 124 and WHV 138) to evaluate its safety and ability to induce high-titer antibodies and strong T-cell responses against diverse HIV-1 subtypes.
