Overview

**PDS0101 + NHS-IL12 + bintrafusp alfa** is an investigational triplet combination immunotherapy for advanced solid tumors, particularly human papillomavirus (HPV)-associated cancers such as cervical cancer. **PDS0101** is a therapeutic cancer vaccine composed of Versamune nanoparticles delivering HPV16 E6/E7 peptides to induce robust CD8+ T-cell responses against HPV oncoproteins. **NHS-IL12** is a tumor-targeted immunocytokine consisting of two interleukin-12 (IL-12) heterodimers fused to the NHS76 monoclonal antibody, which binds DNA/histone complexes in necrotic tumor regions to localize IL-12 delivery, promoting Th1 differentiation, NK/CD8+ T-cell activation, and IFN-γ production while minimizing systemic toxicity. **Bintrafusp alfa** (M7824) is a bifunctional fusion protein combining a transforming growth factor-β (TGF-β) trap (soluble TGF-β receptor II) with an anti-PD-L1 immunoglobulin G1 antibody to simultaneously block immunosuppressive TGF-β signaling and PD-L1/PD-1 interactions in the tumor microenvironment. Developed primarily by PDS Biotechnology for the combination, with NHS-IL12 originating from EMD Serono/Merck KGaA and bintrafusp alfa from Merck KGaA, this regimen aims to synergistically enhance antitumor immunity through vaccination, localized cytokine stimulation, and dual checkpoint modulation.[1][2][3][5][7]
