PEG:CCS13 is a PEGylated single-stranded DNA aptamer developed by D5 Pharma that acts as a selective agonist for the CD200 receptor 1 (CD200R1). CD200R1 is an inhibitory glycoprotein receptor expressed on the surface of various immune cells, including myeloid cells and T-lymphocytes, which plays a key role in downregulating inflammatory responses. By mimicking the natural ligand CD200, PEG:CCS13 activates the receptor to suppress the induction of cytotoxic T-lymphocytes (CTLs) and inhibit the production of pro-inflammatory cytokines. The drug consists of the CCS13 DNA aptamer sequence covalently linked to a 20-kDa polyethylene glycol (PEG) chain at the 5' end to improve its pharmacokinetic properties. Its mechanism of action is distinct from traditional anti-TNF therapies, potentially offering a new therapeutic pathway for patients with rheumatoid arthritis and other inflammatory conditions.
