Overview

Pegargiminase + doxorubicin is a **combination therapy** under investigation for advanced solid tumors, particularly those with argininosuccinate synthase 1 (ASS1) deficiency such as metastatic HER2-negative breast carcinoma. **Pegargiminase (ADI-PEG 20)** is a recombinant, pegylated form of the enzyme arginine deiminase, which depletes extracellular arginine—a nonessential amino acid crucial for the growth of certain cancer cells[3][5][7][9]. **Doxorubicin** is a cytotoxic anthracycline cancer drug that intercalates DNA and inhibits topoisomerase II, leading to DNA damage and apoptosis. Preclinical research demonstrates that arginine depletion potentiates the cytotoxicity of liposomal doxorubicin in arginine-dependent cancer cells by interfering with DNA repair pathways and pyrimidine metabolism, thus synergistically increasing anti-tumor efficacy[3][7][9]. This combination has been studied in a phase 1 trial, showing a manageable safety profile and signs of clinical activity[3][7][6].
