**Pegenzileukin + cetuximab** is an experimental combination of two biologic drugs for cancer therapy. **Pegenzileukin** (SAR444245, THOR-707) is a PEGylated recombinant interleukin-2 (IL-2) analog engineered to selectively activate immune cells without binding to the IL-2 receptor alpha chain, aiming to stimulate T cells and NK cells while reducing activation of regulatory T cells. **Cetuximab** is a chimeric monoclonal IgG1 antibody targeting the epidermal growth factor receptor (EGFR), blocking ligand binding and downstream signaling, leading to inhibition of cell proliferation, increased apoptosis, and immune-mediated tumor cell killing. The combination is being evaluated in advanced solid tumors, based on evidence that combining cytokine-based immunotherapies with EGFR inhibition can enhance anti-tumor immune responses.
