Pegilodecakin + pembrolizumab is a combination therapy that pairs two distinct immunotherapeutic agents. Pegilodecakin (pegylated IL-10) is a first-in-class, long-acting IL-10 receptor agonist that induces oligoclonal T-cell expansion and has anti-inflammatory and CD8+ T-cell stimulating properties. Pembrolizumab is an anti-PD-1 monoclonal antibody immune checkpoint inhibitor. The combination leverages complementary immune-enhancing mechanisms, with pegilodecakin stimulating CD8+ T-cells while pembrolizumab blocks the PD-1 pathway that cancer cells use to evade immune detection.
