peginterferon alfa-2a + imatinib is a combination therapy evaluated for the treatment of chronic myeloid leukemia (CML). It consists of peginterferon alfa-2a, a pegylated recombinant interferon, and imatinib mesylate, a small-molecule tyrosine kinase inhibitor. Peginterferon alfa-2a exerts its effects by binding to the interferon-alpha receptor (IFNAR), inducing antiproliferative, antiviral, and immunomodulatory activities. Imatinib specifically inhibits the BCR-ABL tyrosine kinase, the product of the Philadelphia chromosome (Ph+) that drives the uncontrolled proliferation of myeloid cells in CML. This combination was investigated by Hospices Civils de Lyon, particularly in the SPIRIT trial and Phase II studies, to overcome resistance to imatinib monotherapy and enhance molecular response rates in patients with chronic-phase CML.
