Overview

Peginterferon alfa-2a + ribavirin is a combination therapy used primarily for the treatment of chronic hepatitis C virus (HCV) infection. Peginterferon alfa-2a is a recombinant, pegylated form of human interferon alpha that acts by binding to type I interferon receptors, inducing antiviral and immunomodulatory effects through upregulation of interferon-stimulated genes. Ribavirin is a nucleoside analog with broad-spectrum antiviral activity; its mechanism in HCV therapy includes inhibition of viral RNA polymerase and modulation of immune responses, though the precise mechanism remains incompletely understood. This combination was once standard-of-care for HCV before the advent of direct acting antivirals (DAAs), achieving sustained virologic response rates between 36% and 59% depending on genotype after 48 weeks of treatment[1][2][3][6][8]. Peginterferon alfa-2a is also approved for chronic hepatitis B infection.
